Selective ablation of mast cells or basophils in mice reduces peanut-induced anaphylaxis

چکیده
Background Studies using c-kit mutant mast cell (MC)-deficient mice and antibody-mediated depletion of basophils have suggested that both MCs and basophils can contribute to peanut-induced anaphylaxis (PIA) in mice. However, interpretation of data obtained using such approaches to analyze the contributions of individual effector cells to active anaphylaxis is complicated since mice with mutations affecting c-kit structure or expression have several phenotypic abnormalities in addition to their MC deficiency and basophil-depleting antibodies can also react with MCs.

Peanuts can contribute to anaphylactic shock by activating complement.
BACKGROUND Peanut allergy is the most common food-related cause of lethal anaphylaxis and, unlike other food allergies, typically persists into adulthood. Resistance to digestion and dendritic cell activation by the major peanut allergen Ara h 1 are reported to contribute to its allergenicity. OBJECTIVE We sought to evaluate whether peanut molecules might also promote anaphylaxis through an i...
متن کاملMouse and human neutrophils induce anaphylaxis.
Anaphylaxis is a life-threatening hyperacute immediate hypersensitivity reaction. Classically, it depends on IgE, FcεRI, mast cells, and histamine. However, anaphylaxis can also be induced by IgG antibodies, and an IgG1-induced passive type of systemic anaphylaxis has been reported to depend on basophils. In addition, it was found that neither mast cells nor basophils were required in mouse mod...
متن کاملImpact of CD40 ligand, B cells, and mast cells in peanut-induced anaphylactic responses.
The effector immune mechanisms underlying peanut-induced anaphylaxis remain to be fully elucidated. We investigated the relative contribution of Igs, mast cells (MCs), and FcepsilonRI in the elicitation of anaphylaxis in a murine model. Assessment of peanut hypersensitivity reactions was performed clinically and biologically. Our data show that wild-type (WT; C57BL/6 strain) mice consistently d...
متن کاملHuman FcγRIIA induces anaphylactic and allergic reactions.
IgE and IgE receptors (FcεRI) are well-known inducers of allergy. We recently found in mice that active systemic anaphylaxis depends on IgG and IgG receptors (FcγRIIIA and FcγRIV) expressed by neutrophils, rather than on IgE and FcεRI expressed by mast cells and basophils. In humans, neutrophils, mast cells, basophils, and eosinophils do not express FcγRIIIA or FcγRIV, but FcγRIIA. We therefore...
متن کاملContribution of Classic and Alternative Effector Pathways in Peanut-Induced Anaphylactic Responses
Food allergy affects approximately 5% of children and is the leading cause of hospitalization for anaphylactic reactions in westernized countries. However, the pathways of anaphylaxis in food allergy are still relatively unknown. We investigated the effector pathways of allergic and anaphylactic responses of different strains of mice in a clinical relevant model of peanut allergy. C3H/HeOuJ, C5...
